Abstract

The reversal effect of MDR inhibitors on single-cell drug accumulations has been exploited in order to improve drug sensitivity of the cancer cell. We have applied the same single cell analysis (SASCA) method to measure drug accumulation in acute myeloid leukemia (AML) patient cells by using a microfluidic chip designed to employ dielectrophoresis (DEP) for cell trapping. Generally, we predict a cell is MDR if it has low initial drug accumulation and a substantial fold increase in drug accumulation in the presence of an inhibitor. We have found 78% correlation between our SASCA measurement results and patients' clinical outcomes. Copyright © (2013) by the Chemical and Biological Microsystems Society All rights reserved.
