Abstract
Specific molecular recognition between antibody and receptor on cell membrane plays a central role in biological research. As a main component in the renin-Angiotensin system (RAS), angiotensin II type I receptor (AT1R) mediates the classical biological actions of angiotensin II (Ang II). In this study, we applied atomic force microscopy (AFM) to characterize and quantify binding interaction between anti-AT1R antibody and AT1R on H295R cells. Immunocytochemistry analysis of AT1R was performed on H295R cells to confirm the expression and localization of AT1R. AFM measurement showed the specific unbinding force between anti-AT1R antibody and AT1R was in the range of 50-120 pN. This investigation provides new avenues for mapping AT1R on living cells.
