Abstract
A polymerase chain reaction (PCR) microchip integrated with microheaters and temperature sensors has been designed and fabricated based on micro electromechanic system (MEMS) techniques. The structure of the proposed device is verified by using Finite Element Analysis (FEA) to optimize the thermal performance. The PCR microchip has high heat generation efficiency and heat transfer uniformity in the deoxyribonucleic acid (DNA) amplification. By combining peripheral thermal control system, a temperature fluctuation of ± 1°C, a heating rate of 5°/s, and a cooling rate of 3°C/s have been achieved, and 30 thermal cycles have been finished in 30 minutes. The system has been used for deoxyribonucleic acid (DNA) amplification of GUS gene and good results were obtained from the experiments with much less thermal time. It can be used for DNA sample quick amplification.
